Part:BBa_K2754001:Design
The RRvT, brighter red fluorescent protein, is inspired by the BBa_J04450 biobrick. We wanted to make it comparable to the BBa_J04450 biobrick, and it is therefore designed to have similar conditions. It consists of a lactose induced promoter (Plac BBa_R0010), a ribosome binding site (BBa_B0034), the RRvT sequence and a terminator (BBa_B0015).
The RRvT has very low acid sensitivity according to the article by Wien[1].
The RRvT was synthesized by IDT as a linear sequence containing prefix and suffix. We then ligated it with the pSB1C3 backbone to make it RFC10 compatible. The improved biobrick contain a lactose induced promoter (Plac BBa_R0010), a ribosome binding site (BBa_B0034), the RRvT sequence and a terminator (BBa_B0015). The new biobrick is therefore a composite part. We have not sent the basic part with the RRvT sequence alone into the registry. Only the composite part was tested and sent in to the iGEM registry.
RRvT has an emission wavelength of 583nm and excitation wavelength of 556nm. The protein has a pKa=3.9
